Effect of Endogenous and Exogenous Cholesterol on the Feedback Control of Cholesterol Synthesis.

چکیده
The feeding of cholesterol has been found to result in a prompt suppression of hepatic cholesterol synthesis in all experimental animals thus far studied (1-4). Although the animal body, therefore, has available an effective negative feedback system that can minimize the effects of excessive amounts of exogenous cholesterol, it is not known to what extent this mechanism will respond to hypercholesterolemia of endogenous synthetic origin. The present studies were designed to compare the influence of endogenous and of exogenous hypercholesterolemia upon the feedback control of cholesterol synthesis. Exogenous hypercholesterolemia was produced by feeding chickens a 1 or 2% cholesterol diet for a period of 2 days to 3 weeks. As an experimental model of an endogenous hypercholesterolemic state, use was made of chickens treated for comparable periods with stilbestrol (a,a'-diethylstilbenediol) at a dosage of 5 mg per day, a procedure that is known to produce marked hypercholesterolemia within 2 to 3 days (5). The results of this study demonstrate that the cholesterol feedback system in the chicken is promptly activated by exogenous cholesterol; in contrast, however, the comparable hypercholesterolemia produced by the administration of stilbestrol does not cause a depression in cholesterol synthesis for a period of at least 10 days. Furthermore, the administration of stilbestrol will actually inhibit the feedback response to exogenous

The Effect of Cholesterol and Simvastatin on Passive Avoidance Memory of Streptozotocin-Induced Diabetic Rats
Introduction:Cholesterol has different effects on memory, and diabetes caused adverse effect on cognition. Since there is not enough evidence regarding the effect of cholesterol on memory of diabetic animals, the effects of cholesterol and simvastatin on the passive avoidance memory of diabetic rat were studied. Methods:The study was done on 60 male Wistar rats (180 ± 20g). Diabetes was in...
متن کاملThe interaction of cholesterol absorption and cholesterol synthesis in man.
The total miscible pool of cholesterol in the body is determined largely by the interaction of cholesterol absorption and synthesis. In the present study we have examined the net effects of this interplay in one normal and five hypercholesteremic subjects when various amounts of cholesterol were made available for absorption. Feeding large amounts of cholesterol to the normocholesteremic patien...
متن کاملThe relation between cholesterol absorption and cholesterol synthesis in the baboon.
To determine the relation between cholesterol absorption, total endogenous cholesterol synthesis, and hepatic cholesterol synthesis in a primate, cholesterol synthesis has been studied in biopsies of liver and ileum from normal baboons fed varying amounts of cholesterol and in biopsies of liver from baboons that had been subjected to ileal diversion. In addition, total cholesterol production ra...
متن کاملStudies on the site of the feedback control of cholesterol synthesis.
Despite wide variations in dietary cholesterol, most animals are capable of maintaining the cholesterol of their plasma at relatively constant levels. Insight into the means by which this homeostatis of plasma cholesterol is accomplished was first provided when Gould and associates (3-5), Tomkins, Sheppard and Chaikoff (6) and Frantz, Schneider and Hinkelman (7) independently demonstrated that ...
متن کاملEffect of exogenous cholesterol and dithiothreitol on the activity of human liver microsomal acyl-coenzyme A:cholesterol acyltransferase (ACAT).
Acyl-coenzyme A:cholesterol acyltransferase (ACAT) is the intracellular enzyme responsible for the esterification of cholesterol with long-chain fatty acyl-CoA derivatives and has been implicated in atherosclerosis and gallstone disease. The effects of exogenous cholesterol and dithiothreitol (DTT) on the ACAT activity of human liver microsomes have been determined. Pre-incubation of microsomes...
